To put it simply: When you create a new listing and don't use a saved profile, you must fill out all the fields from scratch. If you create a new listing from a saved profile, you don't have to.
For example, in my main profile I have these saved: 1) A few lines that I always include in the description 2)Shipping information (I always use calculated shipping) 3) Payment info, etc.
That way it makes it much faster to create a new listing. For example when I get to the shipping section all I need to do is enter the weight of the item because the methods, etc are already pre-filled out from my previously saved profile.

A profile is not necessary in order to create a listing and post it to ebay. However, after you have typed the same information a few dozen times in your listings, you will come to realize that profiles can save you lots and lots of repetitive typing, and will therefore stop creating listings until you have spent a bit of time going through each of the profiles and typing in your pertinent information in each of them. You will realize that spending some time getting your profiles done up just so, and then going a step further and creating one or more Master Profiles, will save you loads of time in the future as you create your listings.
